1963年9月出生,安徽人,博士畢業於南京大學生物化學與分子生物學專業,1996年12月至2001年8月先後在美國羅徹斯特大學醫學中心和加拿大多倫多大學附屬病童醫院(SickKids)接受博士後訓練。2001年9月作為教育部“長江學者”特聘教授,受聘為東南大學基礎醫學院遺傳學研究中心主任,擔任博士生導師;領銜申報東南大學第一個生命科學領域博士授權點(遺傳學)獲準;2003年獲準建設“發育與疾病相關基因”教育部重點實驗室,擔任實驗室主任;2004年4月至2009年11月擔任東南大學基礎醫學院院長,2009年12月至2019年12月擔任東南大學生命科學研究院院長,期間完成了東南大學生物學一級學科碩士點、博士點、博士後流動站的建設、協助基礎醫學博士點的獲準;2019年領銜論證成立生命科學與技術學院,獲得東南大學批準,謝維任學院學術委員會主任、東南大學首席教授。

謝維教授先後獲得國家自然科學基金(麵上、重點)、中-加合作項目、國家傑出青年科學基金(2003年)、美國NIH基金(RO3)、國家“973”重大基礎研究項目、國家科技創新2030-“腦科學與類腦研究”(“國家腦計劃”)重大研究項目(首席科學家)等資助。長期從事腦發育的遺傳控製、跨細胞與跨係統信號調節機製和抗體工程靶向藥的研究。發現了自閉症關聯基因參與調控突觸、環路發育;解析了調控社交行為的多條神經環路等;指導創業團隊研發抗體靶向藥,已獲得臨床試驗許可證6張(中、美),其中兩款藥物分別在臨床I、II期實驗。

小鼠早期器官發生過程中完整胚胎的數字化重建

亮點  在早期器官發生過程中,以單細胞分辨率重建完整數字胚胎, 構建中胚層和內胚層譜係的時空圖譜和信號通路圖譜, 識別胚-胚外界麵處的原基決定區 (PDZ), 表征參與器官原基形成的跨胚層信號

總結  早期器官發生是胚胎發育的關鍵階段,其特點是廣泛的細胞命運決定以啟動器官形成,但也極易出現發育缺陷。在此,我們對來自 6 個 E7.5-E8.0 胚胎的 285 個連續切片進行了分析,以單細胞分辨率構建了早期器官發生過程中的完整時空轉錄組和信號通路圖譜。通過開發 SEU-3D,我們重建了數字胚胎,從而能夠在天然的空間環境中研究區域化的基因表達。我們建立了一種基於空間信息的基因-細胞共嵌入方法,係統地構建了內胚層和中胚層衍生物的空間圖譜,並闡明了跨胚層和細胞類型的信號網絡。此外,我們還表征了在胚胎發育第7.75天(E7.75)沿胚胎-胚外界麵形成的原基決定區(PDZ),並揭示了協調的信號通訊促進了心髒原基的形成。總而言之,這種高分辨率的“數字胚胎”為早期器官發生提供了重要的見解,並為研究發育和疾病提供了一個獨特的空間平台。
