Sure.

1. If ask for more because of the cost in locale, do back it up with data. For example, when you say your pay should match the cost of locale, you can say after a comparison, the house market there is twice higher; daycare is 50% higher, etc.

2. If ask for more because of your experience, do back it up with salary data as well.

2. No harm to negotiate. Won't lose the offer right away by asking. They will either stick to the original offer or give you a counter offer.
