  1. Change camera to A mode
  2. Change focus mode to AF-S
  3. Change focus area to Single or Pin-point
  4. Move the focus square to where the eye is
  5. Half-press the shutter button to focus, and hold it in place
  6. Full-press the shutter button to take the picture

For portrait:

Using zoom in 28~70mm range, use f/4 or f/2.8 as the aperture

Using zoom in 70~200mm range, use f/4 or f/5.6 as the aperture

You can always go to f/2.8 or f/2.0 with the longer zoom lens, the background will be really blurred.  Also, the wider the aperture, the thinner the depth of the focus.  With a super long lens, very wide aperture, and relatively short camera-to-subject distance, you may have one eye in focus and the other eye blurred.
