I assume you didn't switch to B2 after your layoff, so you have been out of status in US for a while.
Now you have to take a chance: go back to Canada first, then fly to your work location. Apply for a new TN at the airport. When asked (usually they don't), tell them you are in Canada after your last layoff.
Don't drive your US plate car to a border crossing.
