If your I140 is an employment based one, and you change your job, and because of that, your I485 is denied, then your I140 is useless, unless you go back to work for the employer who sponsor your GC petition. Then you can file your I485 again.

It really depends on the reason I485 got denied. Basicly you can fix it and reapply in most cases.
