If she is in F-1 in US, she has to register before she graduates. OPT or not OPT doesn't matter, because it's the F1 status in question.

If she is out of US but will enter in December for defense, ask her acedemic department whether registration is needed for her defense in fall semseter. Because this is not an immigration issue at all, it's school's policy for student enrollment. And for OPT, international center won't endorse if she is not an enrolled student or not a student just graduated in less than 60 days ago.
