I would file the I485 associated with the EB3. Becuase the job described in your petition is a "future job" that you are going to get after you get your green card, it would be fine if you can explain the job (position) reasonably to the USCIS if they have doubts. In most cases, you will get the I485 approved without any additinal inquiry from uscis.
You don't even know whether your EB2 I140 will be approved of, it make no senses to wait and file the I485 with EB2.
