　　21年前，一股神秘恐怖的邪惡力量奪走了Sam Winchester（Jared Padalecki飾演，代表作《吉爾莫女孩》《恐怖蠟像館》）和Dean Winchester（Jensen Ackles飾演，代表作《超人前傳》《末日黑天使》）的母親。他們的父親John（Jeffrey Dean Morgan飾演，代表作《實習醫生格蕾》）把兄弟倆當成士兵一樣養大，告訴他們黑暗的角落裏躲藏著可怕的惡靈，教導他們辨認史籍中形形色色的鬼怪……並手把手指導他們殺死這些惡魔。
　　然而，Sam不想再過這種動蕩不安的生活，獨自一人前往斯坦福求學。直到有一天哥哥Dean帶著壞消息出現在他家門口：父親在一次獵鬼之旅中失蹤。
　　於是Sam和Dean駕駛著一輛1967年Chevy Impala車，在美國的高速公路網穿梭了整整一年，尋找父親的蹤跡。沿途，他們經曆了各種超自然時間，同時兄弟之間也矛盾不斷。最終，兄弟倆找到了父親，三人一同追蹤殺害母親的黃眼惡魔。經過一場惡戰，父子三人身負重傷，惡魔也趁亂逃跑了。
　　Winchester兄弟的任務仍在繼續。他們走遍全國，遭遇了形形色色的超自然生物――吸血鬼、幽靈、亡魂、甚至死神，而對於大多數人來說，這些東西隻存在於傳說迷信及噩夢之中。
　　無論走到哪兒，兩兄弟從未放棄尋找害得他們家破人亡的黃眼惡魔。在最近一次對抗中，黃眼惡魔提到，他為“Sam和其他像他一樣的孩子”製定了計劃。現在，Sam必須查出其中含義，尋找埋藏在自己及家族過去中的黑暗秘密。
　　本劇華納兄弟電視公司聯合Wonderland聲畫公司出品，McG（代表作《查理的天使》《橘子郡男孩》）、Robert Singer（代表作《狼吻夜驚魂》）及John Shiban（代表作《X檔案》）擔任監製，Eric Kripke擔任編劇兼監製（代表作《惡靈空間》）。
