A few questions to ask:
1. Did it work before?
2. What system are you running?
3. Did you try different USB port on the same computer?
4. Does your USB drive work on other computers?
5. Did you try to see if it works after restarting PC?
6. Did you try "device manager" to see if USB controller is working? If it shows a question mark, you need to reinstall driver.
